When I attach a requirement and click on fail and issue it should go directly to Jira?
Should I integrate Jira first to the project?

There are 2 sides of the integration, one side lets you link between User Stories / Requirement in JIRA to the Requirements in PT - that is the first part of what you wrote.
The second part of the integration, that is independent of the first, is to create and link Issues in JIRA as bugs from your PT runs
for both of them you need to set up the integration first
